使用熒光底物的直接ELISA實驗方案
實驗概要使用熒光偶聯一抗的直接 ELISA 測定的程序。實驗步驟第 1 天：1. 在濾過的 PBS 中稀釋包被抗體后，在每孔中加入 100 μl 進行包被。用封板膜覆蓋酶標板，將酶標板在 4℃下孵育過夜。 第 2 天：2. 將 4 g Block ACE 粉末稀釋于 100 ml 去離子水中，使用其 1:4 稀釋液在每孔中加入 200 μl，在室溫下封閉酶標板 3 小時，封閉期間用封板膜覆蓋酶標板。3. 洗滌酶標板：PBS-T (0.05% Tween20) 250 μl/孔；3x 30 s。在洗滌或抽吸后，將酶標板翻轉置于工作臺的無塵紙上以除去多余的液體。4. 取 100 μl 剛剛在 10% BlockACE 的 PBS-T 溶液中稀釋的標準品或樣品，上樣到酶標板，并在 4℃下孵育過夜，孵育期間用封板膜覆蓋酶標板。提前制備標準品。 在將標準品上樣到酶標板的當天（第二天），現將標準品在例......閱讀全文
